A Deep Dive into PDE5 Inhibition: Innovative Discoveries via Virtual Screening

Background: PDE5 inhibitors have had a surge in popularity over the last decade owing to their efficacy in the treatment of erectile dysfunction, coronary vasculopathy, and pulmonary arterial hypertension. These inhibitors exhibit competitive binding with phosphodiesterase type 5 and inhibit the hydrolysis of cyclic guanosine monophosphate, hence elevating the levels of cGMP in smooth muscle cells and prolonging the duration of an erection. However, due to production costs and side effects, further research is needed to discover new PDE5 inhibitors. background: The field of PDE5 inhibitors has risen in popularity in the past decade because of the success of PDE5 inhibitors in treating erectile dysfunction. Due to the structural identity with cGMP; PDE5 inhibitors competitively bind with PDE5 and limit cGMP hydrolysis, which raises the cGMP level in smooth muscle cells and lengthens the duration of an erection. PDE5 inhibitors were also found to be beneficial for coronary vasculopathy, pulmonary arterial hypertension, and benign prostatic hyperplasia. But, due to the expensive cost of production and unwanted side effects, it is necessary to explore the possibility of the discovery of new PDE5 inhibitors. Characterization and classification of EEG signals evoked by different CAD models

AbstractThe past two decades have witnessed dramatic advancement in computer‐aided design (CAD). However, development of human–computer interfaces (HCI) for CAD have not kept up with these advances. Windows, Icons, Menus, Pointer (WIMP) is still the mainly used interface for CAD applications which limits the naturalness and intuitiveness of the CAD modeling process. As a novel interface, Brain–computer interfaces (BCIs) have great potential in the application of CAD modeling. Utilizing BCIs, the user can create CAD models just by thinking about it in principle, because BCIs provide an end‐to‐end interaction channel between users and CAD models. However, current related studies are mainly limited to the existing BCIs paradigms, while ignoring the relationship between electroencephalogram (EEG) signals and CAD models, which largely increases the cognitive load on the users. In this study, we aimed to explore the potential of using BCI to create CAD models directly independent of the classical BCIs paradigms. For this purpose, EEG signals evoked by six basic CAD models (i.e., point, square, trapezoid, line, triangle, and circle) were collected from 28 participants. After preprocessing and sub‐trial principal components analysis (st‐PCA) of recorded data, the peak, mean and time‐frequency energy features were extracted from EEG signals. By applying the one‐way repeated measures analysis of variance, we demonstrated that there were significant differences among these EEG features evoked by different CAD models. These features from EEG electrode channels ranked by mutual information were then used to train a discriminant classifier of genetic algorithm‐based support vector machine. The empirical result showed that this classifier can discriminate the CAD models with an average accuracy of about 72%, which turns out that EEG based model generation is feasible, and provides the technical and theoretical basis for building a novel BCI for CAD modeling.

G-code generation for deposition of continuous glass fibers on curved surfaces using material extrusion-based 3D printing

Improving the mechanical properties of 3D printed parts produced through a material extrusion-based 3D printer with continuous fibers (carbon, glass, and aramid) has been a focal point for numerous researchers. Given the layered nature of additive manufacturing (AM) processes, wherein parts are built up layer by layer, most studies involve the deposition of continuous fibers onto a 2D surface. Cases involving curved surfaces have employed robots with high degrees of freedom. This research introduces a method for depositing continuous glass fibers onto curved surfaces, implemented on a cost-effective material extrusion-based 3D printer. The presented approach involves G-code modification, the incorporation of a rotating axis for the nozzle, and the application of computer-aided design and manufacturing techniques. Experimental results affirm the efficacy of this method for depositing continuous fibers onto curved surfaces. The developed technique enables the production of free-form composite shells with a thermoplastic matrix and continuous fiber reinforcement. Lastly, through 3D scanning of the printed sample and subsequent comparison with the 3D model, the degree of surface form deviation and tolerance is determined. The maximum deviation identified in this study is 0.1 mm, a tolerable amount considering the inherent characteristics and behaviors of thermoplastic materials (shrinkage and warpage) during production processes.

Hemodynamic-based virtual surgery design of double-patch repair for pulmonary arterioplasty in tetralogy of Fallot

Background and objectiveSurgical correction of pulmonary artery stenosis (PAS) is essential to the prognosis of patients with tetralogy of Fallot (TOF). The double-patch method of pulmonary arterioplasty is usually applied in case of multiple stenosis in TOF patients’ pulmonary artery (PA) and when PAS cannot be relieved by the single-patch method. The surgical planning for the double-patch design remains challenging. The purpose of this study is to investigate the double-patch design with different angulations between the left pulmonary artery (LPA) and the right pulmonary artery (RPA), and to understand postoperative hemodynamic alterations by the application of computer-aided design (CAD) and computational fluid dynamics (CFD) techniques. MethodsThe three-dimensional model of the PA was reconstructed based on preoperative computed tomography imaging data obtained from the patient with TOF. Three postoperative models with different designs of double-patch were created by “virtual surgery” using the CAD technique. Double-Patch 120 Model was created with double patches implanted in the main pulmonary artery (MPA) and the PA bifurcation and without changing the spatial position of PA. The angulation between the LPA and the RPA was defined as θ, which equaled to 120° in Pre-Operative Model and Double-Patch 120 Model. Based on Double-Patch 120 Model, Double-Patch 110 Model and Double-Patch 130 Model were generated with θ equaled to 110° and 130°, respectively. Combined with CFD, the differences of velocity streamlines, wall shear stress (WSS), flow distribution ratio (FDR), and energy loss (EL) were compared to analyze postoperative pulmonary flow characteristics. ResultsThe values of velocity and WSS decreased significantly after virtual surgery. Obvious vortices and swirling flows were observed downstream of the stenosis of RPA and LPA in Pre-Operative Model, while fewer vortices developed along the anterior wall of the expanded lumens of RPA, especially in Double-Patch 110 Model. With the relief of PAS, two relatively higher WSS regions were observed at the posterior walls of RPA and LPA. The maximum WSS values in these regions of Double-Patch 110 Model were lower than those in Double-Patch 120 Model and Double-Patch 130 Model. Furthermore, the FDRs were elevated and the ELs were greatly reduced. It was found that Double-Patch 110 Model with the angulation between the LPA and the RPA equaled to 110° showed relatively better properties of hemodynamics than other models. ConclusionsThe angulation between the LPA and the RPA is an important factor that should be integrated in the double-patch design for TOF repair. Virtual surgery based on patient-specific vascular model and computational hemodynamics can be used to provide assistance for individualized surgical planning of double-patch arterioplasty.

Differentiable Rendering of Parametric Geometry

We propose an efficient method for differentiable rendering of parametric surfaces and curves, which enables their use in inverse graphics problems. Our central observation is that a representative triangle mesh can be extracted from a continuous parametric object in a differentiable and efficient way. We derive differentiable meshing operators for surfaces and curves that provide varying levels of approximation granularity. With triangle mesh approximations, we can readily leverage existing machinery for differentiable mesh rendering to handle parametric geometry. Naively combining differentiable tessellation with inverse graphics settings lacks robustness and is prone to reaching undesirable local minima. To this end, we draw a connection between our setting and the optimization of triangle meshes in inverse graphics and present a set of optimization techniques, including regularizations and coarse-to-fine schemes. We show the viability and efficiency of our method in a set of image-based computer-aided design applications.

Applications of Computer-Aided Drug Design (CADD) tools in the healthcare system are a developing area of research. Implementation of in-silico, structure-based virtual screening techniques in drug discovery to identify potential agonists or inhibitors based upon the binding affinity towards corresponding target proteins is a widely used technique in drug discovery. This advanced technique is used in this study to identify potential candidates from Himalayan pteridophytes possessing antifungal potentials through inhibition of Sec14p, a phosphatidylinositol/phosphatidylcholine transfer protein from Saccharomyces cerevisiae that is complexed with Picolinamide. 3D conformations for the chemical structures of identified phytoconstituents were prepared through Chem Draw 16.0 program for prediction of activity (way2drug), mechanism (PyRx), physicochemical & pharmacokinetics (SWISS-ADME) and toxicological (PROTOX-2) probabilities through in-silico approaches for all selected 180 ligands. BIOVIA discovery studio visualizer was used to generate a 3D and 2D image of the most active 19 among the 180 identified ligands. Comparative studies proposed some highly potential effective and safe compounds from Himalayan pteridophytes that could interfere with fungal cell growth.

Analysis of the application of computer-aided design in industrial product design based on

Abstract The purpose of exploring the application of computer-aided design technology in industrial product design is to realize the diversification of industrial product design and meet the personalized customization needs of customers. Starting from computer-aided technology, this paper illustrates the optimization of CAD design by using a genetic algorithm and constructs a bipolar progressive interactive genetic algorithm using bipolar progressive ranking and fuzzy fitness jointly. Genetic algorithm genotypes in CAD process product design are defined, and a comparative evaluation experimental analysis of industrial product design is performed with the AR-IGA algorithm. Regarding the number of iterations, the average number of iterations for the AR-IGA algorithm to complete the color-matching design of process products among ten testers was 9.34, which was 36.42% lower than that of the TIGA algorithm. The average evaluation elapsed time of the AR-IGA algorithm was 246.9s, which was 24.12% less than the average evaluation elapsed time of the TIGA algorithm. This shows that computer-aided technology optimized by genetic algorithms can effectively improve the efficiency of industrial product design and satisfy more customers to realize personalized design simultaneously.

A morphological study of fivefold Islamic geometric patterns using formal grammar for computer-aided design applications

This paper presents a morphological study of Islamic geometric patterns (IGP) and their role in application of formal grammar in computational modeling of IGPs. Through a comprehensive literature review and data collection, we analyze the morphological properties of these patterns using techniques such as geometric transformations, pattern classification, and symmetry analysis. Based on our findings, we explore how these properties can be used in constructing a formal grammar through string rewriting system for a CAD application. Building on a study on the potential of a string rewriting system for modeling IGPs, the current research suggests an update to the previous system and introduces a new morphological structure for IGPs. The new method has an expanded sample set and is tested on a class of 5-fold star patterns with 12 members and demonstrates successful development. The results are implemented in a Grasshopper add-on, providing a flexible platform to generate the patterns through strings and to control their parameters. This tool opens up new possibilities to bridge traditional patterns with contemporary technologies and make them more accessible. Furthermore, it contributes to the preservation of IGPs as a significant cultural and architectural heritage, while also advancing the evolution of these patterns to a new and contemporary generation.

Generalized Affine Equivalence Checking of Boolean Functions via Reachability Analysis

We consider the problem of checking the generalized affine equivalence of two given Boolean functions. This problem arises in various computer-aided design (CAD) and cryptographic applications, such as circuit synthesis and Reed-Muller codes. Based on the theory of affine group acting on the Boolean functions, we define the coefficient spaces and transition relations, and transform the checking problem into reachability analysis of finite state machines. Two methods are proposed to check the affine equivalence of Boolean functions using Binary Decision Diagrams (BDDs) and Property Directed Reachability (PDR), respectively. Both methods can check affine equivalence of bent and semi-bent functions, which state-of-the-art methods can hardly handle. Furthermore, existing methods only consider the case of affine equivalence, while our methods can handle the generalized affine equivalence of subspaces of Boolean functions. In the application of circuit synthesis, our methods can significantly reduce the size of the library compared to Boolean matching. To classify Boolean functions up to the generalized affine equivalence, we propose a method to obtain a complete classification based on BDDs. In the experiments, we have successfully applied our methods to some examples that can hardly be solved by using the previous methods, thus validating the effectiveness of our methods.

B-rep Matching for Collaborating Across CAD Systems

Large Computer-Aided Design (CAD) projects usually require collaboration across many different CAD systems as well as applications that interoperate with them for manufacturing, visualization, or simulation. A fundamental barrier to such collaborations is the ability to refer to parts of the geometry (such as a specific face) robustly under geometric and/or topological changes to the model. Persistent referencing schemes are a fundamental aspect of most CAD tools, but models that are shared across systems cannot generally make use of these internal referencing mechanisms, creating a challenge for collaboration. In this work, we address this issue by developing a novel learning-based algorithm that can automatically find correspondences between two CAD models using the standard representation used for sharing models across CAD systems: the Boundary-Representation (B-rep). Because our method works directly on B-reps it can be generalized across different CAD applications enabling collaboration.

Eco-friendly UV-spectrophotometric methods employing magnetic nano-composite polymer for the extraction and analysis of sexual boosters in adulterated food products: Application of computer-aided design.

Solid phase extraction (SPE) techniques based on computationally designed magnetic-based multi-targeting molecular imprinted polymer (MT-MIP) and combined with UV spectrophotometric approaches provide advantages in the examination of counterfeit samples. The current work describes an innovative and sustainable methodology for the simultaneous determination of tadalafil (TAD) and dapoxetine hydrochloride (DAP) in counterfeited honey and instant coffee aphrodisiac products utilizing a SPE exploiting MT-MIP. Besides, an innovative UV spectrophotometric method capable of resolving TAD in its pharmaceutical binary mixtures with DAP was developed. A novel computational approach was implemented to tailor the synthesis and design of the MT-MIP particles. Herein; we applied a newly developed UV spectrophotometric method which was based on Fourier self-deconvolution (FSD) method coupled with isoabsorptive point for determination of TAD and DAP in pharmaceutical dosage form. Besides we applied a SPE process based on the MT-MIP designed particles, assisting in the analysis of both drugs in counterfeited food samples. The SPE process and the UV spectroscopic methodology were assessed regarding their greenness using the pioneering GAPI, AGREEprep and AGREE greenness assessing tools. The synthesized MT-MIP particles were characterized by scanning electron microscopy and energy-dispersive X-ray spectroscopy. The suggested spectrophotometric methods revealed a wider linear concentration range of 2-50 µg/mL with lower limits of detection in the range of 0.604 -0.994 µg/mL. Additionally, the suggested method demonstrated the utmost sensitivity and eco-friendliness for their target in its mixed dosage form and counterfeited food products. The SPE process and the developed analytical UV spectroscopic methodology were validated as per the ICH guidelines, and were found to be suitable for overseeing some counterfeiting activities in commercially available honey and instant coffee aphrodisiac products. An SPE based on MT-MIP magnetic-based polymer assisted with UV-spectroscopic method was successfully developed for analysis of tadalafil and dapoxetine hydrochloride in different matrices.

Outstanding productions of peroxymonosulfuric acid combining tailored electrode coating and 3D printing

In this work, the combination of a tailored design of electrode and electrochemical cell to produce peroxymonosulfuric acid (PMSA), also known as Caro's acid, is evaluated. For the electrode, a customized coating of BDD was deposited onto a niobium substrate. This electrode exhibited a good reproducibility and efficiency in the production of PMSA, thus promoting direct oxidation of sulfuric acid into Caro's acid and preventing the action of scavengers. The electrochemical cell was manufactured after the application of computer-aided design (CAD) combined with computational fluid-dynamics (CFD) simulation and 3D printing. The prototype, with an area of 25sqcm and a designed flowrate of 48 L/h, has been characterized with the evaluation of mass transfer coefficients and RDT curves, which demonstrate an outstanding performance with mass transport coefficients over 3·10−5 m s−1 without turbulence promoter. The cell equipped with the electrodes was tested in the production of Caro's acid enabling a remarkable production of 200 mM of peroxymonosulfuric acid in discontinuous operation mode with a coulombic efficiency (57.2 %) and energy Efficiency (10.67 mmolA−1 h−1). These values outperform the previous state-of-the-art works done in the field for undivided cells operated in discontinuous mode. The results obtained were successfully fitted to a phenomenological model for the generation of Caro's acid.

Automated layout of modular high-rise residential buildings based on genetic algorithm

The conventional design process of modular high-rise residential buildings (MHRBs) heavily relies on the experience of designers, leading to concerns about design efficiency and quality. Although some methods have been suggested for generating floor plans, there is a scarcity of studies focusing on the automated layout of MHRBs. This paper utilizes the genetic algorithm (GA) to automatically generate MHRB layouts. The approach consists of two stages within the GA framework: the first stage handles the layout design of modular flats, while the second stage is dedicated to the layout design of the traffic core. To facilitate the process, computer-aided design (CAD) drawings are automatically generated using the Autodesk CAD application programming interface, utilizing the information acquired from the proposed approach. Additionally, a website is developed to enable interaction with designers, and two examples are provided to demonstrate the effectiveness of the proposed approach.

Building on prior methods used to aid correlation of MRI results and whole-mount pathology findings, we have developed an inexpensive and easy to implement means with which 3D-printed molds can be created for any prostate to enable targeted ex-vivo sampling of biopsy-proven cancer lesions seen on MRI. The potential applications for a platform that can reliably sample living prostate cancer tissue are numerous, including the ability to facilitate future cancer research. METHODS: STL files of the prostate boundary and target lesions are exported from the UroNav fusion biopsy system. A custom macro was developed for use with SolidWorks (Dassault Systèmes, France), a computer-aided design application. This creates a material-optimized mold around the prostate with needle guides that allow for targeted sampling of up to 4 regions of interest. The guides are customizable in terms of location and dimensions to accommodate different biopsy needles. The parts are then exported and 3D-printed. Following prostate extraction at time of radical prostatectomy, the seminal vesicles are removed. The prostate is then placed into the mold, and samples are taken by inserting a biopsy needle into a guide up to the predetermined depth. The prostate is then passed to pathology for standard processing, while the live tissue in the biopsy cores is available for a variety of purposes. RESULTS: The resulting platform allows for a prostate of any size or morphology to be precisely sampled as long as there is a pre-acquired MRI dataset. The prostate mold allows for accurate sampling of target cancer lesions and is optimized in size for each prostate to allow for minimal material waste. CONCLUSIONS: This novel platform provides the unique ability to obtain fresh prostate cancer tissue. The utility of this is not necessarily limited to prostate cancer alone but may be of use in other solid-organ malignancies with targetable lesions as well. Research on the Application of Computer-Aided Industrial Design and 3D Printing Technology: A Comprehensive Perspective Analysis

With the continuous development and popularization of computer-aided industrial design technology and 3D printing technology, the integration of the two is also receiving more and more attention. Starting from the basic principles and development history of computer-aided industrial design technology and 3D printing technology, this paper introduces the advantages and current situation of the integration of the two, and focuses on exploring the integration of computer-aided industrial design and 3D printing technology in product design, process analysis, and manufacturing process optimization. Finally, the development trend and future prospects of integration application are summarized.

Application of computer-aided design and 3D-printed template for accurate bone augmentation in the aesthetic region of anterior teeth

PurposeTo explore the outcomes of bone augmentation in the aesthetic zone of the anterior teeth using computer-aided design and a 3D-printed template.MethodsTen patients with severe bone defects in the aesthetic zone of anterior teeth were included in the study; CT data were collected before surgery. The design of the osteotomy line in the bone defect area was determined under computer simulation. The position parameters and osteotomy line of the free bone were determined via virtual surgery. A 3D-printed template was prepared to guide the accurate placement of the bone graft. Reexamination was conducted to evaluate the position of the bone graft immediately after the operation and the resorbed capacity of the bone graft before implant restoration.ResultsThe position of the bone graft was consistent with the preoperative design. The amount of bone graft resorbed was within the acceptable range three months after the operation, and the effect of implant restoration was satisfactory.Clinical significanceUse of computer-aided design and a 3D-printed template can be an effective approach for accurate bone augmentation in the aesthetic zone of the anterior teeth.

Application of Computer Aided Design (CAD) and Flat Techniques in Teaching Pattern Drafting by Clothing Lecturers in Universities in South East, Nigeria

The purpose of the study was to investigate the application of computer aided design (CAD) and Flat techniques in teaching pattern drafting by clothing lecturers in Universities in South East, Nigeria. The population for the study was 48 lecturers. The entire number of Clothing lecturers in the above three universities were used because of the manageable size. A questionnaire was utilized for data collection (0.94 Cronbach’s alpha). Data collected were analyzed using mean and standard deviation. The major findings of the study showed that some of the challenges in the application of CAD and Flat instructional techniques in teaching pattern drafting include lack of skills on the part of the teachers, insufficient numbers of computers and insufficient technical support for teachers, among others. Orientation of teachers on the importance of computer aided design instruction, using blended approach in teaching, and use of another pattern drafting technique among others are strategies to overcoming the challenges of CAD and Flat technique of pattern drafting instruction by Clothing Lecturers. Among the recommendations made was that curriculum planners and lecturers should make effort to adopt the use of CAD as one of the instructional techniques in teaching pattern drafting.

But an excellent curve with parameters need to eliminate unnecessary cusps and inflection points, so shape analysis is needed for some specific curves. We have already shown many excellent properties of the DTB-like curves and DT B-spline-like curves. Thus the shape features like convexity, loops, inflection points, and cusps of the DTB-like curves, and DT B-spline-like curves are further discussed in this paper. And the necessary and sufficient conditions for the existence of shape features of the corresponding DTB-like curves and DT B-spline-like curves are given. And the shape features distribution all be generalized into tables and diagrams, which are useful for industrial design. In addition, the effect of shape parameters on the shape features diagram and its adjustment ability to the shape of corresponding curves are analyzed, respectively. The work in this paper enables users to determine how to set parameter values so that the generated curve could be a global convex or local convex curve, has a required inflection point, or eliminate the unnecessary one, and could be adjusted to another aimed shape.
